Active siteThe active site is similar to that of other cysteine-type deubiquitinating enzymes, and a Cys/His catalytic dyad has been identified by site-directed mutagenesis. A glutamine N-terminal to the active site cysteine probably represents the residue that helps form the oxyanion hole during catalysis, because Ala or Glu mutants at this position are catalytically inactive. A residue C-terminal to the active site histidine that orientates the imidazolium ring has not been identified (Abdul Rehman et al., 2016).
Activities and specificitiesOnly Lys48-linked polyubiquitin chains are cleaved. Ubiquitin molecules are released one at a time (Abdul Rehman et al., 2016).
Molecular structureMembers of the MINDY family uniquely amongst deubiquitinating enzymes contain a ubquitin-binding domain known as an MIU (motif interacting with ubiquitin) motif, which is a helical motif that binds mono-ubiquitin (Penengo et al., 2006). <#FAM118B#C115.002#> contains an EF hand structure inserted in the peptidase unit. The structure of <#MINDY-1#C115.001#> has been solved (Abdul Rehman et al., 2016).
