ElaD (Escherichia coli) and SseL (Salmonella enterica serovar Typhimurium) were discovered to be deubiquitinylating peptidases in 2007 (see bleow).

Active siteActive site Cys residues have been identified by mutagenesis (Catic et al., 2007; Rytkonen et al., 2007). The active site His is recognised by similarity to clan CE peptidases (see the Alignment for clan CE).
Activities and specificitiesElaD and the similar SseL from Salmonella enterica serovar Typhimurium hydrolyse ubiquitinyl bonds. Recombinant ElaD hydrolysed ubiquitinyl aminomethylcoumarin and showed other reactions characteristic of a ubiquitinyl hydrolase (Catic et al., 2007). A fusion protein containing SseL hydrolysed mono- and polyubiquitin substrates in vitro with a preference for Lys63-linked ubiquitin chains, but also cleaving at Lys-48 (Rytkonen et al., 2007).
InhibitorsElaD (but not its C312S form) reacted with a potential inhibitor in which a Michael acceptor was added to the C-terminus of ubiquitin (Catic et al., 2007).
