Active siteThe sequence of DUBA deubiquitinylating peptidase contains an 'OTU-like' domain (as described by Makarova et al., 2000) that is found in a number of families in clan CA (Kayagaki et al., 2007). Predicted active site residues are Asp221, Cys224, His334 (see the Alignment), of which the Cys was shown by mutagenesis to be essential for activity.
Activities and specificitiesDUBA selectively cleaved the lysine-63-linked polyubiquitin chains on tumor necrosis factor receptor-associated factor 3 (TRAF3), resulting in its dissociation from a downstream signaling complex.
