Active siteActive site residues have not been determined for any member of this family, and the active site triad is hypothetical. As can be seen from the Alignment for clan SC, the pattern of conserved glycines around the proposed active site serine and histidine residues in family S37 is consistent with that of other families in this clan.
Activities and specificitiesThe peptidase from Streptomyces mobaraensis has been shown to activate the transglutaminase precursor by removal of a leading tri- or tetrapeptide (Umezawa et al., 2004). Calcium ions enhance activity (Zotzel et al., 2003).
InhibitorsPS-10 peptidase (S37.001) is inhibited by the generic serine peptidase inhibitor PMSF (Umezawa et al., 2004).
Molecular structureNo tertiary structure has been determined for any member of the family. Peptidases in family S37 are synthesized with signal peptides (Umezawa et al., 2004).

Biological functionsPS-10 peptidase processes the transglutaminase precursor from Streptomyces mobaraensis. Processing occurs in two stages: a metallopeptidase (M04.017) or the serine peptidase SAM-P45 (S08.069) removes the bulk of the propeptide leading to an active enzyme (Zotzel et al., 2003); PS-10 peptidase then removes an N-terminal tri- or tetrapeptide (Umezawa et al., 2004).
