Name and HistoryADAMDEC1 is an unusual member of the ADAM subfamily because it lacks a disintegrin and an ADAM-CR domain, and the third zinc ligand is aspartic acid rather than histidine (within the motif HEXXHXXGXXD) (Bates et al., 2002).

SpecificityDespite the third zinc ligand being aspartic acid, ADAMDEC1 is proteolytically active and cleaves the bait region of alpha2-macroglobulin. Replacing the aspartic acid with histidine dramatically increases cleavage (Lund et al., 2013). Cleavage of carboxymethylated transferrin shows a restricted specificity for Leu in P1' (Lund et al., 2015).
Inhibitor commentsBatimastat is inhibitory. TIMPs do not inhibit the wild type peptidase, but TIMP-3 inhibits the Asp362His mutant (Lund et al., 2015).
StructureThe sequence consists of a prodomain and a metallopeptidase domain. The human sequence has an unpaired cysteine residue in the catalytic domain (Lund et al., 2015).
PhysiologyADAMDEC1 was initially thought to be involved in the immune response because it was present in mature, follicular dendritic cells and germinal centres in the spleen (Mueller et al., 2001) and macrophages (Fritsche et al., 2003). However, it is also found in uterine stromal cells (Baran et al., 2003) and transcripts are highly expressed in pulmonary sarcoidosis (Crouser et al., 2009).
